以乳及乳制品为原料,利用高效液相色谱法检测其中硫氰酸钠的含量。实验主要针对各类乳制品各成分指标的差异,建立不同提取净化提取方法。样品经离心沉淀蛋白后,利用碱性氧化铝小柱去除氨基酸、维生素等。以磷酸盐缓冲液:乙腈=95∶5(pH=7.00±0.02)作为流动相,在218nm下,通过Durashell HLP C18色谱柱(4.6*250mm,5μm),柱温30℃,流速为0.8mL/min,采用紫外检测器分析样品。结果显示:该方法对乳及乳制品中硫氰酸钠有较高的提取率,回收率均在80%以上。
Abstract:The content of sodium thiocyanate was determined by high efficiency liquid chromatography in milk and dairy products. Different purification and extraction method was established according to the different component of all kinds of dairy products. The protein of sample was handled with precipitation and centrifugation,using alkaline alumina column removal of amino acids,vitamins and other materials. Phosphate buffer: acetonitrile = 95 ∶ 5( pH = 7. 00 0. 02)as the mobile phase,in 218 nm,through the Durashell HLP C18( 4. 6* 250 mm,5μm),the column temperature is30℃,flow rate of 0. 8mL /min,using UV detector analysis sample. The method had high extraction rate of sodium thiocyanate in milk and dairy products,the recovery rate was above 80%.
