本研究建立了复配营养强化剂中维生素D3超高效液相色谱检测方法。样品加入氨水,于55℃超声波水浴中处理,冷却后加入三氯甲烷、乙醇提取维生素D3,离心分离,取上清液用0.22μm有机系滤膜过滤,采用ACQUITY UPLC HSS T3色谱柱分离,流动相为甲基叔丁基醚、甲醇、0.1%甲酸溶液,梯度洗脱,紫外检测器采集数据,外标法定量。对方法进行验证,维生素D3在其质量浓度范围内线性关系良好,标准曲线的R2 > 0.99,回收率为91.0%~102.0%,相对标准偏差为2.29%~3.52%,检出限为0.02μg/g。该方法快速、准确,适合各类复配营养强化剂中维生素D3的测定。
Abstract:An ultra-high performance liquid chromatography(UHPLC) method was established for the determination of vitamin D3 in compound nutritional fortifiers. Samples were treated with ammonia in an ultrasonic water bath at 55 ℃. After cooling,vitamin D3 was extracted using a mixture of trichloromethane and ethanol. Chromatographic separation was performed on an ACQUITY UPLC HSS T3 column using a mobile phase of methyl tert-butyl ether,methanol,and 0.1% formic acid. Detection was conducted using a UV detector and quantified using the external standard method. The method demonstrated excellent linearity(R2>0.99),with recoveries ranging from 91.0% to 102.0% and relative standard deviations(RSD) between 2.29% and 3.52%. The limit of detection(LOD) was 0.02 μg/g. This method was rapid,accurate,and suitable for the routine analysis of vitamin D3 in various compound nutritional fortifiers.
